FERTILIZATION OF GOAT OOCYTES MATURATED IN TWO CULTURE MEDIA

Abstract


The objective of this study was to evaluate the effect of two culture media on the in vitro maturation (IVM) and fertilization (IVF) rates of goat oocytes. Cumullus oocyte complexes (COC’s) were obtained from ovaries of postmortem goats. In experiment I (EXP I), 361 COC´s were randomly distributed for maturation in TCM-199 (n=171) and HECM-9 (n=190) media. Both media were supplemented with LH and FSH (10µg/ml) and 50µg/ml gentamicina. COC´s were cultivated at 38.5 °C, 5% CO2. Twenty-seven hours post-maturation, the COC’s were washed in PBS and stained with Hoescht 33342 to evaluate the nuclear maturation. In experiment II (EXP II) 667 CCO´s were distributed in TCM-199 (n=323) and HECM-9 (n=344), and 30 oocytes of each treatment were evaluated as in EXP I, the rest were co-incubated with 2x104 buck spermatozoa in TBM medium + 0.03% BSA to evaluate the fertilization rate after 36 h. In EXP I, the nuclear maturation rate was higher (p<0.05) in HECM-9 (60.5%), respect to TCM-199 (38.6%). In vitro maturation (IVM) and in vitro fertilization (IVF) rates in the EXP II were higher (p<0.05) in the HECM-9 (84.8% and 19.18%, respectively) than in TCM-199 media (62.7% and 9.59%, respectively).The HECM-9 is able to support the in vitro maturation and fertilization of goat oocytes.
